ABSTRACT

Objective To establish a submandibular gland(SMG) cell model of radiation injury, and to observe the effect of compatibility of Fructus Mume(Wumei) and Radix Glycyrrhizae(Gancao) on the proliferation of the modeled cells. Methods SMG cells were primarily cultured in vitro for the experiment. After immunofluorescence identification and growth curve analysis, SMG cells were given one-time exposure to X electron-ray at dose of 0.1, 0.2, 1, 2, 4 Gy respectively, and then were cultured with Wumei-Gancao solution at the concentration of 500, 50, 5, 0.5 μg/mL separately. Methyl thiazolyl tetrazolium (MTT) was used to determine the survival rate of SMG cells exposed to different doses of X electron-ray and proliferation vitality of SMG cells treated with different concentrations of Wumei-Gancao solution for 24 h. Results The survival rate of SMG cells was decreased gradually with the increase of X electron-ray dose. When the concentrations of Wumei-Gancao solution were in the range of 50 ~ 0.5 μg/mL, the cell proliferation rate showed an increasing trend with the increase of drug concentration, the effect being dose-dependent. When the solution concentration arrived to 500 μg/mL, the cell proliferation rate was declined. Conclusion The SMG cell model of radiation injury has been established successfully under the conditions of 0.1 Gy of one-time exposure, 9 Mev of energy at dose rate 3 Gy/min, hop count at 12 MU and source-skin distance being 100 cm. The concentration of the solution with the compatibitity ratio of Wumei to Gancao being 3:1 has certain effects on repairing the modeled cells.

ABSTRACT

OBJECTIVE: To investigate the difference of chemical constituents between Radix Glycyrrhizae and processed Radix Glycyrrhizae products and to analyze the influence of processing on the chemical constituents. METHODS: Using HPLC, the chemical composition of licorice, licorice fried with honey and fried licorice were analyzed. The contents of glucose, sucrose, D-fructose and 5-hydroxymethyl-2-furaldehyde were determined. RESULTS: The proportions of the main chemical components were changed after Radix Glycyrrhizae et Rhizoma was processed. 5-HMF emerged and the contents of glucose and fructose increased. The effects of different processing methods on the chemical constituents were different. CONCLUSION: The changes in the ratios between components, which result from heating processing and honey processing and the addition of honey, may be the material basis for the enhanced pharmacological action of Radix Glycyrrhizae et Rhizoma after it is stir-baked with honey. It could be helpful to study the principle of stir-baking Radix Glycyrrhizae et Rhizoma with honey.

ABSTRACT

Objective To analyze the relationship between clinical features of infantile cough after infection and microelements in peripheral blood, and to observe the therapeutic of treating this cough with Milkvetch Root and Radix Glycyrrhizae. Methods 75 infantile patients were randomly recruited into a therapeutic group and a reference group. 39 patients in the therapeutic group were treated with Huangqi Zhusheye and Tablete Glycyrrhiza, while 36 patients in the reference group were treated with Azithromycin. The therapeutic effects were evaluated after the treatment. Meanwhile,microelements in peripheral blood of the 75 patients were assayed and made a contrast to 40 healthy children. Results The contents of Fe, Zn, and Mg in peripheral blood of the 75 patients dropped on different levels, comparing to the healthy children.The therapeutic effects of the therapeutic group was much better than the reference group with significant difference (P<0.05).Liver and renal functional lesion was unseen in both groups after the treatment. Conclusion Infantile past-infection cough may related with the decrease of Mg, Fe, and Zn. Combined usage of Milkvetch Root and Radix Glycyrrhizae clinically has good therapeutic effects to this disease.

ABSTRACT

OBJECTIVE:To establish the quality standard for Xiaoer Zhike oral liquid(XZOL).METHODS:The Herba Ephedrae,Pericarpium Citri Reticulatae and Radix Glycyrrhizae in XZOL were identified by TLC.The chlorogenic acid in the liquid was determined by HPLC,The mobile phase was methanol-water(10∶90)with a flow rate of1ml/min,the detection wavelength was254nm.RESULTS:The Herba Ephedrae,Pericarpium Citri Reticulatae and Radix Glycyrrhizae in XZOL could be identified on TLC chromatograms.The linear range of chlorogenic acid was0.2?g~1.2?g(r=0.9996),the average recov?ery was96.67%,RSD was1.04%.CONCLUSION:The method is simple,reliable and accurate.The method can be used for quality control of XZOL.

ABSTRACT

Objective To establish the quality standard of Fengsui Pills. Methods The qualitative analysis of Fructus Amomi and Radix Glycyrrhizae in Fengsui Pills was carried out by TLC,and the content of berberine hydrochloride,an effective component of Cortex Phellodendri in Fengsui Pills was determined by HPLC. Results The chromatographic sports of Fructus Amomi and Radix Glycyrrhizae were identified without the interference of negative control. A linearity of berberine hydrochloride was obtained from 0.272?g to 1.360 ?g with a good correlation (r=0.999 9,n=5).The average recovery was 99.01 %and the RSD was 0.98%. Conclusion The qualitative and quantitative methods established in this study are simple and feasible,accurate and reproducible,and can be used as the quality control standard for Fengsui Pills.

ABSTRACT

Objective To study HPLC fingerprints of Radix Glycyrrhizae from different habitats, develop the method of hierarchical cluster analysis by using the results of fingerprints, and discuss regional effect on accumulation of active ingredients of Radix Glycyrrhizae. Methods HPLC Fingerprint analysis method of active ingredients was developed and the method had been evaluated. Based on the standardized result of peak area of glycyrrhizin, liquiritin, liquiritigenin, isoliquiritin, and isoliquiritigenin, seventeen batches of Radix Glycyrrhizae were classified and identified using hierarchical cluster analysis. Results The methodological evaluation showed that this method had a good repeatability. The amounts of glycyrrhizin and flavones in Radix Glycyrrhizae from various habitats are different. Conclusion Accumulation of active ingredients of Radix Glycyrrhizae has some relativity with habitats.

ABSTRACT

Objective To establish the HPLC fingerprints of Radix Glycyrrhizae. Methods The RP-HPLC method was used, Zorbax SB C_ 18 column (250 mm?4.6 mm, 5 ?m) was employed; the acetonitrile-1.2% acetic acid (gradient elution) was used as mobile phase, analytic time was 70 min, and detective wavelength was at 254 nm. Results The HPLC fingerprints of Radix Glycyrrhizae were set up. The result showed that 38 peaks were common in different sources. The results of method validation met technical standard of fingerprints. Conclusion The method is stable and reliable with a good reproducibility and provides a reference standard for the quality control of Radix Glycyrrhizae.

ABSTRACT

Object To develop an immunoassay method of species-specific protein for Radix Glycyrrhizae (RG) identification. Methods The crude drug of RG as a model was studied in this paper,the antigen of RG protein (RGP) was screened by Western-blot analysis using anti-RG serum. After species-specific protein,RGP was purified,and rabbit antiserum against RGP was prepared,the rabbit IgG of anti-RGP was isolated and biotinylated,then the sandwich enzyme-linked immuosorbent assay (ELISA) was developed. Results The method was only specific to RG,which was harvested from various areas or species,and showed excellent sensitivity and reproducibility. Conclusion The result suggests that the immunoassay method using specific antigen of RGP as a detection objective be a new potential for the unequivocal identification and quality control of RG.

ABSTRACT

Object To study the effects of SINI TANG (SNT) on the rat aortic rings pre-contracted by high K + and phenylephrine (Phe). Methods The effects of SNT on the aortic rings in the presence of 60 mmol/L KCl and Phe (1?10 -9 -1?10 -4 mmol/L) were observed and t heir me chanisms were studied after treatment with Propranolol (Pro, 3?10 -6 mmol/ L) and Bay K8644 (BK, 1?10 -5 mmol/L) as tool drugs. Results SNT inhibited the contraction induced by cumulative Phe and decreased the maximum tension (T max ); Pro couldn't influe nce the effects of SNT. SNT attenuated the amplitude of contractile effect of hi gh K +; BK couldn't reverse the effects of SNT. Conclusion SNT can shift the dose-response curve to the right and decrease the T max . It shows that SNT is a kind of noncompe titive antagonism. SNT decreases the effect of high K + against contraction of the artery. BK, a L-type Ca 2+ channels activator, couldn't recover the inhibition induced by SNT. The results suggest that SNT inhibit ? 1 recep tor, while calcium channel may not be involved in attenuating the effect of SNT on high K +-induced contraction.

ABSTRACT

Objective To set up the quality standard of Wanbile Wan. Method s Fructus Psoraleae, Radix Puerariae, Radix paeoniae Alba and Radix Glycyrrhiza e Preparatae in Wanbile Wan were identified by TLC .Isopsoralen was determined b y TLC scanning. The chromatographic conditions were: high performance silica gel G plate as lamellated plate, n- hexane - ethyl acetate (8 ∶ 2)being the exp ander, serrated scanning with single- wavelength reflection, beam stricture of 0.4? 0.4 mm, linear parameter SX=3 and scanning wavelength at 300 nm. Results The TLC spots were highly specific, clear and concentrated without interferen ce of the presence of negative controls. Isopsoralen showed a good linearity wit hin the range of 0.064~ 1.536 ? g, r=0.9980, average recovery rate was 100.7 % , and RSD was 2.0% .Conclusion This method can be used for the quality co ntrol of Wanbile Wan.

ABSTRACT

AIM: To set up a new method to analyze IR fingerprint spectra, which was in line with the cha racteristic of traditional Chinese medicine. METHODS: In this paper. two new indexes, common peak ratio and variant peak ratio, were applied and its values were calculated by means of the sequential analysis, in which each sample's IR fingerprint spectra was set up and the common peak ratio sequences were arranged in order of size in comparison with other samples. RESULTS: The method could distinguish the water extract of Radix Glycyrrhizae of different varieties and different areas. CONCLUSION: The dual index sequential analysis enables us to distinct two or more herb's IR fingerprint.

ABSTRACT

AIM: To investigate the effect of glycyrrhizin(GZ) and glycyrrhefinic acid(GA) on reducing the toxicity of Radix Aconiti Laterlis(RAL) and its mechanisms of action. METHODS: Radix Aconiti Laterlis(RAL),RAL with Radix Glycyrrhizae(RG),RAL with GZ,RAL with GA were boiled for 30 min,respectively.HPLC was used to measure the concentrations of aconitine and hypaconitine in the different decoctions.Meanwhile,the acute toxicity of the above decoctions was determined by oral and intraperitoneal administration in mice respectively. RESULTS: By oral administration,the acute toxicity of the decoctions of RG with RAL and GE with RAL was obviously lower than that of RAL.The acute toxicity of the decoction of GA with RAL was slightly reduced when compared with that of RAL.By intraperitoneal administration,the acute toxicity of GA with RAL was obviously higher than that of RAL.RG significantly restrained the dissolution of alkalines contained in RAL into the decoction.But GA and GZ increased the dissolution of the alkalines. CONCLUSION: By oral administration,RG could reduce the acute toxieity of RAL.Restraint on the dissolution of the toxic alkalines contained in RAL into the decoction may be in part the reasons.But GA and GZ are not the effective components reducing the dissolution of the toxic alkalines.They may combine with the toxic alkalines,postpone the absorption of the combined compounds through gastrointestinal tract,and reduce the acute toxicity of RAL by oral administration.

ABSTRACT

OBJECTIVE:To study the infuence of mixed decoction of Cortex Phellodendri,Radix Glycyrrhizae and Ramu?lus Loranthi on extraction rate of berberine.METHODS:The extraction amount from Cortex Phellodendri,Radix Glycyrrhizae and Ramulus Loranthi mixed decoction was determined by HPLC.The HPLC conditions were:Hypersil BDS C 18 column;mobile phase,acetonitrile-33mmol/L KH 2 PO 4 -triethylamine(20∶72∶0.1);detecting wavelength,345nm;column temperature25℃;flow fate1.0ml/min.RESULTS:Decoction of Cortex Phellodendri,Radix Glycyrrhizae and Ramulus Loranthi in combi?nation obviously decreased the extraction rate of berberine.CONCLUSION:Cortex Phellodendri should be extracted separately in formulating extract technic for preparations containing above-mentioned herbs.
